Decoding the genetic link between diabetes, hyperlipidemia, and IBD

Previous epidemiological studies have shown a close association between type 1 diabetes mellitus (T1DM), hyperlipidemia, and inflammatory bowel disease (IBD), but a causal relationship has not been established. . The authors of this paper used Mendelian randomization (MR) analysis to assess the causal relationship between T1DM and hyperlipidemia with IBD.

A two-sample MR study was conducted utilizing accessible genome-wide association study data from public sources and selecting appropriate instrumental variables that adhered to principles of MR analysis. The main technique utilized was the inverse variance weighting method, which was complemented by additional methods such as MR-Egger regression, weighted median, simple mode, weighted mode, MR pleiotropy residual sum and outlier approach . Based on MR analysis, genetically determined T1DM had no causal association with IBD or IBD subtypes.

These findings were consistent across all supplementary methods used. Furthermore, genetically determined hyperlipidemia had no causal relationship with IBD or IBD subtypes, even after increasing the number of instrumental variables used. This study supports the concept that there is no causal relationship between T1DM and IBD and hyperlipidemia and IBD, which is inconsistent with most observational studies.
